چکیده

Previous reports from this laboratory demonstrated the synthesis of hyaluronic acid by a cell-free preparation obtained from a strain of Group A streptococcus. The enzymic activity which sedimented at 105,000 x g, but not at 10,000 x g, required the addition of uridine diphospho-N-acetylglucosamine, uridine diphosphoglucuronic acid, and Mg++ for the formation of hyaluronic acid (l-3). Since hyaluronic acid occurs in the capsule of streptococci, this system affords an opportunity for the localization of synthesis of a high molecular weight polymer found external to the cell. The availability of “phage-associated lysin” capable of solubilizing the cell wall of Group A streptococcus (4) facilitated the preparation (5) of protoplast membranes used in this study. This paper reports the synthesis of hyaluronic acid by protoplast membrane preparations that are relatively free of nucleic acids and cell wall material. A preliminary report of this work has appeared (6). Additional information regarding the properties of the hyaluronic acid-synthesizing system is presented.

The Synthesis of Type III Pneumococcal Capsular Polysaccharide from Uridine Nucleotides by a Cell-free Extract of DipZococcus

In 1953, Bernheimer (1) showed that suspensions of resting Type III pneumococci could synthesize capsular polysaccharide from glucose or from certain other monosaccharides in a medium containing magnesium, potassium, and phosphate ions. Although a considerable amount of polysaccharide was synthesized from glucose, little or none was formed when glucose was replaced by sodium glucuronate or by s...

Prokaryotic expression, purification and immunogenicity analysis of CpsD protein from Streptococcus iniae

Streptococcus iniae is a major cause of serious bacterial infections in both fish and human beings. Capsular polysaccharide (CPS) of S. iniae is vital to evade phagocytic clearance of the host and serves as an important protective antigen of S. iniae infection in aquatic animals. The CpsD gene was determined to be highly conservative in capsule polysaccharide operon. Prokaryotic expression of t...

Hyaluronic acid capsule is a virulence factor for mucoid group A streptococci.

Mucoid strains of group A Streptococcus have been associated with recent outbreaks of acute rheumatic fever. The mucoid colony morphology of these strains is a result of abundant production of capsular polysaccharide, which is composed of hyaluronic acid. To study the role of the hyaluronic acid capsule in virulence, we derived an acapsular mutant from a mucoid strain of group A Streptococcus b...

The biosynthesis of hyaluronic acid by group A Streptococcus. VI. Biosynthesis from uridine nucleotides in cell-free extracts.

Previous studies have demonstrated that both the glucosamine and glucuronic acid moieties of hyaluronic acid synthesized by Group A streptococci are derived from glucose without previous scission of the glucose molecule (1). Shortly after the discovery by Caputto et al. (2) of the role of uridine diphosphoglucose in the epimerization of glucose, Park (3-5) noted the accumulation of uridine nucl...

Definition of a bacterial virulence factor: sialylation of the group B streptococcal capsule.

Sialylation of bacterial capsules has been proposed as an important virulence factor for several species of encapsulated pathogens, including group B Streptococcus. We have constructed a transposon mutant strain of type III group B Streptococcus that expresses a capsular polysaccharide differing from the wild type only in that the mutant strain's capsule lacks sialic acid. The mutant polysaccha...
